目的:研究冠心平对冠脉结扎导致心肌缺血犬心肌细胞凋亡及Bcl-2、Bax蛋白表达的影响。方法:36头Beagle犬,分成假手术组,心肌缺血模型组,心可舒组,小、中及大剂量冠心平组。前两组分别给予空白胶囊,其他各组每日两次口服相应剂量的药物,连续给药10天,10天后进行冠状动脉结扎实验。结扎180min后取心肌切片保存,检测心肌细胞凋亡指数(AI),并用免疫组化法检测心肌组织Bcl-2、Bax蛋白的表达。结果:与模型组相比,心可舒组及冠心平小、中、大剂量组的心肌凋亡指数水平均有所下降,Bcl-2蛋白表达增强,Bax蛋白表达减少,与模型组比较均具有统计学意义(P<0.01)。结论:冠心平具有减轻缺血心肌细胞凋亡,提高Bcl-2蛋白表达,降低Bax蛋白表达,增加Bcl-2/Bax比值的作用。提示冠心平对缺血犬具有抑制心肌细胞凋亡,减轻细胞凋亡对缺血心肌的损伤的作用。
Objective: To study the effects of Guanxinping on myocardial apoptosis and expression of Bcl-2 and Bax protein in myocardial ischemia-induced myocardial ischemia in dogs. Methods: 36 Beagle dogs were divided into sham operation group, myocardial ischemia model group, XK group, small, medium and high dose Guanxintong group. The first two groups were given blank capsules, the other groups were orally administered the corresponding dose of drug twice a day for 10 days, 10 days after coronary artery ligation experiments. After ligation for 180 min, the myocardial slices were taken and the cardiomyocyte apoptosis index (AI) was detected. The expression of Bcl-2 and Bax protein in myocardium was detected by immunohistochemistry. Results: Compared with the model group, the cardiac apoptotic index of XK, SMC group were decreased, the expression of Bcl-2 protein and Bax protein were decreased, compared with the model group With statistical significance (P <0.01). Conclusion: Guanxinping can relieve the apoptosis of ischemic cardiomyocytes, increase the expression of Bcl-2, decrease the expression of Bax and increase the ratio of Bcl-2 / Bax. These results suggest that Guanxinping can inhibit cardiomyocyte apoptosis in ischemic dogs and reduce the effect of apoptosis on ischemic myocardium.
