目的 分析肺癌患者Th1和Th2两类细胞因子的基因表达与转录因子T bet和GATA3表达的相关性 ,探讨黄芪和 (或 )川穹嗪对Th2型肿瘤的干预作用。方法 利用RT PCR技术检测 42例肺癌患者人外周血单个核细胞 (PBMC)中IFNγ、IL 2、IL 4、IL 6、IL 10、T bet、GATA3以及肺癌组织特异性mRNACK19和CEA的表达状况 ;体外加入黄芪和 (或 )川穹嗪后 ,观察IFNγ、IL 4、T bet和GATA3表达的变化情况。结果 42例肺癌患者IFNγ、IL 2、IL 4、IL 6和IL 10的表达频率依次为 4/42 ,5 /42 ,2 7/42 ,2 4/42和 31/42 ;表达强度依次为 0 .10 9,0 .116 ,0 .318,0 .30 6和 0 .372。转录因子T bet和GATA3的表达频率为 6 /42和 34 /42 ,表达强度为 0 .139和 0 .35 9,而且CEA和CK19mRNA阳性的患者中 ,T bet的表达频率和表达强度较低 ,GATA3的表达频率较高 ,表达强度也相应提高 ,表明黄芪和 (或 )川穹嗪能干预这种现象。结论 肺癌患者Th2类细胞因子群谱的高表达与T bet表达降低 ,GATA3表达增高有关 ,黄芪和 (或 )川穹嗪能降低Th2类细胞因子和GATA3的表达 ,提高Th1类细胞因子和T bet的表达。
Objective To analyze the correlation between the expression of Th1 and Th2 cytokines and the expression of T bet and GATA3 in patients with lung cancer, and to explore the intervention effect of astragalus and (or) strychnine on Th2 tumors. Methods RT - PCR was used to detect the expression of IFNγ, IL 2, IL 4, IL 6, IL 10, T bet, GATA3 and tissue specific mRNACK19 and CEA in peripheral blood mononuclear cells (PBMCs) from 42 patients with lung cancer. After adding Astragalus and / or Chuanmazine in vitro, the changes of IFNγ, IL 4, T bet and GATA3 expression were observed. Results The expressions of IFNγ, IL 2, IL 4, IL 6 and IL 10 in 42 lung cancer patients were 4/42, 5/42, 27/42, 24/42 and 31/42, respectively .10 9,0 .116, 0 .318,0 .30 6 and 0 .372. The expression frequencies of T bet and GATA3 were 6/42 and 34/42, respectively. The expression intensity was 0.139 and 0.359, and the expression frequency and expression of T bet were lower in CEA and CK19 mRNA positive patients. GATA3 expression is higher, the expression intensity is also increased accordingly, indicating that astragalus and (or) chrysogen can interfere with this phenomenon. Conclusion The high expression of Th2 cytokines in patients with lung cancer is related to the decrease of T bet and the increase of GATA3 expression. Astragalus and / or chuvynmazine can reduce the expression of Th2 cytokines and GATA3 and increase the Th1 cytokines and T bet expression.
