目的探讨低氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)在门静脉高压食管静脉曲张大鼠食管的局部表达及意义。方法 60只雄性Wistar大鼠随机分成实验组和对照组各30只。实验组入腹后行门静脉一步法结扎加左肾上腺静脉结扎制备大鼠食管静脉曲张模型;对照组行假手术。在模型建立后第7d、14d及28d后测量门静脉压力、食管下段黏膜下血管数目与血管横截面积,并采用免疫组化SABC法检测和比较HIF-1α两组大鼠食管下段中的表达。结果实验组各时相的门静脉压力、食管下段黏膜下血管数目与血管横截面积均较相应对照组明显升高(P<0.01),对照组内各时相门静脉压力、食管下段黏膜下血管数目与血管横截面积之间存在差异(P<0.05)。实验组术后14d及28d大鼠食管下段HIF-1α的表达较对照组明显增强(P<0.01),术后7d与对照组比较无明显差异(P>0.05)。结论在门静脉高压食管静脉曲张大鼠中,HIF-1α对食管静脉曲张的形成和发展起促进作用。
Objective To investigate the expression of hypoxia inducible factor-1α (HIF-1α) in the esophagus of esophageal varices rats with portal hypertension and its significance. Methods Sixty male Wistar rats were randomly divided into experimental group and control group. In the experimental group, one-step ligation of the portal vein and ligation of the left adrenal vein were performed to establish the model of esophageal varices in rats. The control group was sham-operated. The portal vein pressure, the number of submucosal blood vessels and the cross-sectional area of blood vessels in the lower esophagus were measured at 7d, 14d and 28d after model establishment. The expression of HIF-1alpha in the lower esophagus was detected by immunohistochemical SABC method. Results The portal venous pressure, the number of submucosal blood vessels and the cross-sectional area of blood vessels in the lower esophagus were significantly higher than those in the corresponding control groups (P <0.01), the portal venous pressure in each phase in the control group, the submucosal blood vessels in the lower esophagus And vascular cross-sectional area (P <0.05). The expression of HIF-1α in esophageal lower segment of experimental group at 14d and 28d after operation was significantly higher than that of control group (P <0.01). There was no significant difference between the experimental group and the control group on the 7th day after operation (P> 0.05). Conclusion HIF-1α promotes the formation and development of esophageal varices in portal hypertension rats with esophageal varices.
