目的研究粉防己碱(TET)对人膀胱移行细胞癌BIU-87细胞株裸鼠移植瘤的作用机制。方法-14只裸鼠右后腿背侧皮下接种BIU-87移植瘤细胞后随机分为磷酸盐缓冲液(PBS)对照组(7只)和25μg·mL-1TET治疗组(7只)。接种1个月后开始,每3 d一次于肿瘤局部注射给予PBS或TET,每3 d测量1次肿瘤大小;RT-PCR检测Bcl-2、Bax mRNA表达;Western-blot分析survivin、Capseae-3蛋白表达。结果 TET对人膀胱癌BIU-87细胞裸鼠移植瘤的生长具有抑制作用,抑制率达66.6%。RT-PCR检测结果显示,与PBS对照组相比,TET能明显下调移植瘤Bcl-2 mRNA的表达,上调Bax mRNA的表达(P<0.05);Western-blot结果显示经TET处理后survivin蛋白出现明显下调,而Capseae-3蛋白的表达上调明显(P<0.05)。结论 TET能够抑制人膀胱癌BIU-87细胞裸鼠移植瘤的生长,其作用的分子机制与下调Bcl-2、survivin和上调Bax、Capseae-3基因表达有关。
Objective To study the mechanism of tetrandrine (TET) on human bladder transitional cell carcinoma BIU-87 cells in nude mice transplanted tumor mechanism. Methods-14 nude mice were inoculated subcutaneously with BIU-87 transplanted into the dorsal hind paw and were randomly divided into phosphate buffered saline (PBS) control group (7) and 25μg · mL-1TET treatment group (7). The mice were injected with PBS or TET once a day for 3 days after tumor inoculation. The tumor size was measured every 3 days. The expression of Bcl-2 and Bax mRNA was detected by RT-PCR. The expressions of survivin, Capseae-3 Protein. Results TET had an inhibitory effect on the growth of human bladder cancer BIU-87 cells in nude mice with the inhibition rate of 66.6%. The results of RT-PCR showed that compared with PBS control group, TET significantly down-regulated the expression of Bcl-2 mRNA and up-regulated the expression of Bax mRNA (P <0.05). Western-blot showed that survivin protein was detected after TET treatment Obviously down-regulated, while the expression of Capseae-3 protein was significantly up-regulated (P <0.05). Conclusion TET can inhibit the growth of human bladder cancer BIU-87 cells in nude mice. The molecular mechanism of TET is related to the down-regulation of Bcl-2, survivin and up-regulation of Bax and Capseae-3 gene expression.
