本研究在资源库中发现了一个大粒品种,暂被命名为big grain 3(bg3).该品种的千粒重明显高于常规水稻品种.通过遗传分析表明该大粒性状为数量性状.与R28相比,bg3的籽粒长和宽分别增加了36.45%和25.81%,呈极显著增加.利用F2代群体中与R28相同粒型的植株提取DNA用于定位主效基因BG3,最终将BG3定位于第二染色体长臂的SSR引物RM1342和RM6之间,其遗传距离分别为0.027 cM和0.033 cM.通过测序比对发现,Os02947280的第二个外显子有两个碱基的变化,使得BG3的编码提前终止.该结果为后续的研究提供了依据.“,”This study found a big grain variety in the repository,which was temporarily named big grain 3 (bg3).The 1 000-grain weight of bg3 was obviously heavier than that of conventional rice varieties.Genetic analysis showed that this big grain trait was quantitative trait.The grain length and width of bg3 significantly increased by 36.45% and 26.81% compared with R28.DNA was extracted from plants of the F2 population with the same grain size as R28 for locating gene BG3,the result of which showed that the main effect gene BG3 was located between the SSR primer RM1342 and RM6 in the long arm of second chromosome,with the genetic distance of 0.027 cM and 0.033 cM,respectively.Sequence comparison discovered that two bases in the second exon of OsO2g47280 were changed,resulting in the early termination of BG3 coding.The results laid a foundation for subsequent research.
