目的:探讨淫羊藿含药血清对绝经后骨质疏松症大鼠MSCs成脂分化的干预作用。方法:SPF级SD雌性大鼠,共15只。无菌条件下分离培养骨髓间充质干细胞,传至3代。将所获的细胞随机分为空白组、成脂诱导组、成脂诱导西药组、成脂诱导中药高剂量组、成脂诱导中药中剂量组、成脂诱导中药低剂量组共6组,每组8孔。通过淫羊藿高、中、低剂量含药血清以及尼尔雌醇含药血清配合成脂诱导剂分别对MSCs进行干预,并与空白组、成脂诱导剂组进行对比;观察各组干预后7,14d成脂分化情况。结果:诱导第7d,与空白组比较,中药各组油红染色均呈显著性差异(P<0.05);成脂诱导组呈极显著性差异(P<0.01);与成脂诱导组比较,成脂诱导西药组油红染色有极显著性差异(P<0.01),但成脂诱导中药组呈显著性差异(P<0.05)。诱导第14d,各组与空白组比较均呈显著性或极显著性差异(P<0.05,P<0.01);与成脂诱导组比较,成脂诱导中药高剂量组油红染色呈显著性差异(P<0.05),成脂诱导西药组有极显著性差异(P<0.01)。结论:成脂诱导中药高剂量组及成脂诱导西药组均能抑制MSCs成脂分化,但短时间内高剂量淫羊藿含药血清功效略逊于西药尼尔雌醇。
Objective: To investigate the intervention effect of Epimedium medicated serum on adipogenic differentiation of MSCs in postmenopausal osteoporosis rats. Methods: SPF SD female rats, a total of 15. Bone marrow-derived mesenchymal stem cells were isolated and cultured under aseptic conditions and passed on to passage 3. The cells were randomly divided into blank control group, adipogenic induction group, adipogenic induction western medicine group, adipogenic induction high-dose traditional Chinese medicine group, adipogenic-induced traditional Chinese medicine middle dose group, and adipogenic induction low-dose traditional Chinese medicine group. Group 8 holes. MSCs were intervened by high, middle and low dose of serum containing Epimedium, and serum containing nilestriol with adipogenic agent respectively, and compared with blank group and adipogenic induction group. After the intervention 7,14 d adipogenic differentiation. Results: On the 7th day after induction, compared with the blank group, there was a significant difference (P <0.05) in the oil red staining of the Chinese medicine groups; the difference was significant between the adipogenic induction group and the adipogenic induction group (P <0.01) There was a significant difference (P <0.01) between adipose-induced western medicine group and oil-stained Chinese herbal medicine group (P <0.05). On the 14th day after induction, compared with the blank group, there was a significant or very significant difference (P <0.05, P <0.01). Compared with the adipogenic group, the adipose-induced high dose group of traditional Chinese medicine showed a significant difference (P <0.05). There was a significant difference in adipogenic induction group (P <0.01). Conclusion: The adipogenic differentiation of MSCs induced by adipogenic induction of Chinese medicine high-dose group and adipogenic-induced western medicine group can inhibit the adipogenic differentiation of MSCs, but the effect of high dose Epimedium serum is less than that of western medicine nylestriol in a short time.
