目的研究天智颗粒对慢性脑缺血模型神经元特异性烯醇化酶(NSE)表达的影响。方法随机分为对照组、假手术组、模型组和天智治疗组,用剪断双侧颈总动脉的方法建立慢性脑缺血模型,用天智颗粒(5g/kg)治疗1月后采用三等分 Y 型电迷宫和免疫组化方法,观察、比较各组大鼠学习记忆能力和 NSE 表达的差异。结果治疗1月后,治疗组与模型组相比.学习记忆能力显著增加(P<0.05),假手术组和对照组大鼠皮质神经元无明显 NSE 表达,模型组表达明显,治疗组 NSE 表达较模型组减弱,具统计学意义,均 P<0.05。结论天智颗粒可提高慢性脑缺血大鼠学习记忆能力,并可以减弱慢性脑缺血诱发的 NSE 表达,这是天智颗粒治疗血管性痴呆和慢性脑缺血的一种可能机制。
Objective To study the effect of Tianzhi Granule on the expression of neuron-specific enolase (NSE) in chronic cerebral ischemia model. METHODS: Patients were randomly divided into control group, sham operation group, model group, and Tianzhi treatment group. Chronic cerebral ischemia models were established by cutting bilateral common carotid arteries. After treatment with Tianzhi granules (5 g/kg) for one month, trisections were used. Y-type electric maze and immunohistochemistry methods were used to observe and compare the differences of learning and memory abilities and NSE expression among rats in each group. Results After 1 month of treatment, the learning and memory ability of the treatment group was significantly higher than that of the model group (P<0.05). There was no significant NSE expression in the cortical neurons of the sham-operated group and the control group. The expression in the model group was obvious, and the expression of NSE in the treatment group was significantly increased. Compared with the model group, it was statistically significant (P<0.05). Conclusion Tianzhi granules can improve the learning and memory abilities of rats with chronic cerebral ischemia, and can reduce the NSE expression induced by chronic cerebral ischemia. This is a possible mechanism of Tianzhi granules in the treatment of vascular dementia and chronic cerebral ischemia.
