目的:研究卵巢上皮性恶性肿瘤中肾上腺髓质素(AM)的表达及其对ERK活性的影响,探讨其作用机制及临床意义。方法:应用RT-PCR、MTT比色法和蛋白质印迹法对卵巢上皮性恶性肿瘤(30例)、卵巢良性肿瘤(10例)和正常卵巢组织(10例)的AM表达情况及AM、AM受体阻断剂(AM22-52)对卵巢癌细胞增殖和ERK活性的影响进行分析。结果:AM在卵巢上皮性恶性肿瘤中的半定量结果为0.948±0.150,与正常卵巢及卵巢良性肿瘤比较,差异有统计学意义,P=0.042。其表达量与术后病理分级相关,与手术病理分期无关;MTT法检测显示,AM能促进卵巢癌细胞增殖的作用,随着作用浓度和作用时间的增加这种促进作用也随之增强;蛋白质印迹法检测,随着AM作用浓度的增加,磷酸化的ERK1/2的表达随之增高,反之,随着AM受体阻断剂(AM22-52)浓度的增加,磷酸化的ERK1/2的表达随之降低,差异有统计学意义,P<0.05。结论:卵巢癌组织中AM高表达,其能通过MAPK/ERK信号转导途径在卵巢癌的发生中起作用。
Objective: To investigate the expression of adrenomedullin (AM) and its effect on the activity of ERK in epithelial ovarian cancer and to explore its mechanism of action and its clinical significance.
