对诱导1d、3d、5dLAK细胞作ANAE、ACP、POX及PAS细胞化学染色,同时采用LDH释放法对LAK细胞的细胞毒性进行了检测。结果显示,随诱导天数的延长,LAK细胞胞质中ANAE、ACP和PAS的反应物逐渐增多、增强,阳性率也逐渐增高,不同天数组间有显著性差异(P<0.001)。POX均为阴性反应。细胞毒性试验证实,随诱导天数的延长LAK细胞杀伤活性逐渐增强。这表明LAK细胞的细胞化学活性与其细胞毒性密切相关,为了解LAK细胞的激活状态提供了形态学依据。
The cytotoxicity of LAK cells was detected by chemiluminescence staining of ANAE, ACP, POX and PAS on day 1, day 3 and day 5, and LDH release assay. The results showed that the number of reactants of ANAE, ACP and PAS in LAK cell cytoplasm gradually increased with the induction of lengthening, and the positive rate also increased gradually. There was a significant difference between different days (P <0.001). POX are negative reactions. Cytotoxicity test confirmed that with the induction of prolonged LAK cell killing activity gradually increased. This indicates that the cytochemical activity of LAK cells is closely related to its cytotoxicity and provides a morphological basis for understanding the activation state of LAK cells.
