目的探讨消渴颗粒剂治疗糖尿病肾病的机制。方法用高浓度葡萄糖刺激系膜细胞,加入中药含药血清后,采用逆转录多聚酶链反应(reversetranscriptasepolymerasechainreaction,RT-PCR)方法观察中药对高糖刺激后系膜细胞TGF-β1mRNA表达的影响。结果高浓度葡萄糖可刺激系膜细胞TGF-β1mRNA表达,而中药可抑制TGF-β1mRNA的过度表达。结论消渴颗粒剂可抑制高糖引起的TGF-β1mRNA过度表达,这可能是其防治糖尿病肾病的关键。
Objective To investigate the mechanism of Xiaoke granules in treating diabetic nephropathy. METHODS: Mesangial cells were stimulated with high concentration of glucose, and serum containing traditional Chinese medicine was added. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to observe the effect of Chinese herbal medicine on the expression of TGF-β1 mRNA in mesangial cells stimulated by high glucose. Results High concentration of glucose stimulated the expression of TGF-β1 mRNA in mesangial cells, while Chinese medicine could inhibit the overexpression of TGF-β1 mRNA. Conclusion Xiaoke granule can inhibit the overexpression of TGF-β1 mRNA induced by high glucose, which may be the key to prevent and treat diabetic nephropathy.
