目的:阐明软肝缩脾丸对肝纤维化大鼠肝脏胶原基因调节及蛋白表达的影响,探索肝纤维化发生的分子生物学机理。方法:采用免疫组化和原位杂交技术分别测定各组肝纤维大鼠治疗前后肝脏Ⅰ、Ⅲ型胶原的基因调节和蛋白表达。结果:CCl_4模型组大鼠肝脏Ⅰ、Ⅲ型胶原mRNA及蛋白水平明显高于治疗组。正常组大鼠肝组织无1例阳性。结论:软肝缩脾丸治疗可显著降低肝纤维化大鼠肝脏Ⅰ、Ⅲ型胶原的基因和蛋白表达,从而发挥其抗肝纤维化作用。
Objective: To elucidate the effects of Ruangan Shupi Pill on hepatic collagen gene regulation and protein expression in hepatic fibrosis rats, and to explore the molecular biological mechanism of hepatic fibrogenesis. Methods: Immunohistochemistry and in situ hybridization techniques were used to determine the gene regulation and protein expression of hepatic type I and type III collagen in hepatic fibers of rats before and after treatment. Results: The mRNA and protein levels of collagen I and III in liver of CCl_4 model group were significantly higher than those in the treatment group. There was no positive liver tissue in normal rats. Conclusion: The treatment with Ruangan Shupi Pill can significantly reduce the expression of the type I and type III collagen proteins in liver of rats with hepatic fibrosis, thus exerting its anti-hepatic fibrosis effect.
