目的观察三氧化二砷(AS_2O_3)对大肠癌细胞株SW620的生长抑制作用并初步探讨其机制.方法大肠癌细胞株SW620用不同浓度AS_2O_3处理.①MTT法观察不同浓度AS_2O_3作用不同时间后对SW620细胞生长的影响;②增殖细胞核抗原(PCNA)单抗LSAB免疫组化染色-流式细胞仪检测 PCNA阳性细胞百分比.结果①AS_2O_3明显抑制大肠癌细胞株SW620细胞增殖,其作用随AS_2O_3浓度增加和作用时间延长而增加;②随AS_2O_3浓度增加,PCNA阳性细胞百分比逐渐下降(p<0. 05)结论二氧化二砷可明显抑制大肠癌细胞株 SW620的生长,调控 DNA合成及细胞增殖的 PCNA蛋白合成降低可能是 AS_2O_3的作用机制之一.
Objective To observe the growth inhibitory effect of arsenic trioxide (AS_2O_3) on human colorectal cancer cell line SW620 and to explore its mechanism. Methods Colorectal cancer cell line SW620 was treated with different concentrations of AS_2O_3. ①MTT method was used to observe the effects of different concentrations of AS_2O_3 on the growth of SW620 cells at different time; ② PCNA monoclonal antibody LSAB immunohistochemical staining - flow cytometry to detect the percentage of PCNA positive cells. Results ① AS_2O_3 significantly inhibited the proliferation of SW620 cells. The effect of AS_2O_3 increased with the increase of AS_2O_3 concentration and the duration of action. ② As the concentration of AS_2O_3 increased, the percentage of PCNA positive cells decreased gradually (p <0.05). Conclusion Arsenic dioxide Inhibition of growth of human colon cancer cell line SW620 and the decrease of synthesis of PCNA which regulate DNA synthesis and cell proliferation may be one of the mechanisms of action of AS_2O_3.
