目的探讨Toll样受体(Tolllikereceptor,TLR)在人孕早期母胎免疫耐受中的作用。方法应用流式细胞术对28例非妊娠妇女,29例孕早期正常妊娠妇女,25例孕早期妊娠低孕酮妇女,16例孕早期妊娠低孕酮经过黄体酮治疗妇女,15例孕早期自然流产妇女的外周血淋巴细胞、单核细胞、粒细胞TLR2、TLR4检测进行分析。结果妊娠对照组单核细胞TLR2与TLR4的表达(20.37%±14.37%;52.58%±13.65%)显著高于自然流产组(11.25%±13.69%;31.91%±20.03%)(P<0.05),却低于非妊娠对照组(29.99%±16.98%;60.69%±8.68%)(P<0.05);妊娠对照组淋巴细胞、粒细胞TLR2表达(1.83%±0.83%;2.64%±1.34%)低于自然流产组(3.60%±2.37%;5.55%±3.99%)(P<0.05),TLR4无明显差异(P>0.05),与非妊娠对照组相比,淋巴细胞、粒细胞TLR2、TLR4的表达均无显著性差异(P>0.05)。低孕酮妊娠组与妊娠对照组、黄体酮组相比,单核、淋巴、粒细胞TLR2、TLR4表达均无显著性差异。结论孕早期妇女外周血先天性免疫相关受体TLR2、TLR4表达的改变可能与妊娠免疫耐受有关。
Objective To investigate the role of Toll-like receptor (TLR) in immune tolerance of human fetuses in early pregnancy. Methods 28 cases of non-pregnant women, 29 cases of normal pregnant women in the first trimester, 25 cases of low progesterone in the first trimester of pregnancy, 16 cases of low progesterone in the first trimester of pregnancy after progesterone treatment of women, 15 cases of early pregnancy natural Abortion women’s peripheral blood lymphocytes, monocytes, granulocytes TLR2, TLR4 detection were analyzed. Results The expressions of TLR2 and TLR4 in monocytes of pregnant control group were significantly higher than that of spontaneous abortion group (20.25% ± 13.69%; 31.91% ± 20.03%, P <0.05) (29.99% ± 16.98%; 60.69% ± 8.68%) (P <0.05). The expression of TLR2 in lymphocytes and granulocytes in pregnant control group was significantly lower than that in non-pregnant control group (1.83% ± 0.83%; 2.64% ± 1.34% There was no significant difference in TLR4 between spontaneous abortion group (3.60% ± 2.37% and 5.55% ± 3.99%, P <0.05). Compared with non-pregnant control group, TLR4, TLR2, TLR4 There was no significant difference (P> 0.05). Compared with pregnant control group and progesterone group, the expression of TLR2 and TLR4 in mononuclear, lymphoid and granulocyte of low progesterone pregnancy group had no significant difference. Conclusion The changes of innate immune-related receptors TLR2 and TLR4 in peripheral blood of pregnant women in early pregnancy may be related to immune tolerance in pregnancy.
