利用等电聚焦聚丙烯酰胺凝胶电泳方法,分析了大白菜、甘兰、绿菜花、甜椒中8个常用杂交种及其亲本(共24份材料)的种子水溶蛋白电泳图谱。结果表明,等电聚焦电泳技术分辨率高,电泳条带多而且细,各材料蛋白谱带都在20条左右,信息量大,利于进行深入细致的分析。8个杂交品种(F1)与其亲本间蛋白电泳图谱都有明显区别,并以此做为检验纯度的依据,进一步建立了这8个杂交种的纯度检验技术,同时在田间进行了验证性测定,二者鉴定结果相符,证明了本方法的可靠性。
The isoelectric focusing polyacrylamide gel electrophoresis was used to analyze the electrophoresis patterns of seed water soluble protein of eight commonly used hybrids and their parents in 24 Chinese cabbage, cabbage, green cauliflower and sweet pepper. The results showed that the resolution of isoelectric focusing electrophoresis was high, the electrophoresis band was more and more fine, the protein bands of each material were about 20, and the amount of information was large, which would facilitate in-depth and detailed analysis. The electrophoretogram of 8 hybrids (F1) and their parents were significantly different, and as a basis for testing the purity, further established the purity test of these 8 hybrids, at the same time in the field of confirmatory determination, The results of the two are in good agreement, which proves the reliability of this method.
