目的:观察复方绞芪方对非酒精性脂肪性肝炎(NASH)大鼠的影响,探讨其防治NASH的机制。方法:采用高脂高糖饮食喂养12周诱导SD大鼠建立NASH大鼠模型,同时用不同剂量复方绞芪方治疗,观察其对NASH大鼠肝指数、血清肝功能、血脂、血糖的影响;常规HE染色观察肝组织的病理改变并计算NAFLD活动度积分(NAS)。结果:模型组大鼠的肝指数、血清ALT、AST、GLU、LDL-C水平较正常组明显增加;而血清HDL-C水平显著下降;同时NAS计分明显升高。应用复方绞芪方进行干预后,大鼠NAS计分较模型组大鼠显著降低,肝功能、血糖明显改善,血清CHOL、TG、LDL-C水平不同程度下降同时HDL-C明显升高。结论:复方绞芪方能改善肝脏病理组织学变化,明显降低NAFLD活动度积分(NAS),这可能是其治疗高脂高糖饮食诱导的非酒精性脂肪性肝炎的作用机制之一。
OBJECTIVE: To observe the effect of compound strangulation prescription on nonalcoholic steatohepatitis (NASH) rats and to explore its mechanism of prevention and treatment of NASH. Methods: NASH rats were induced by high fat and high sugar diet for 12 weeks to induce NASH rat model. At the same time, different doses of compound prescription of Shuqian Fang were used to observe the effect of NASH on liver index, serum liver function, blood lipid and blood glucose. Routine HE staining was used to observe the pathological changes of liver tissue and calculate NAFLD activity score (NAS). Results: The liver index, the levels of serum ALT, AST, GLU and LDL-C in model group were significantly higher than those in normal group, while the levels of serum HDL-C were significantly decreased. Meanwhile, NAS score was significantly higher. Compared with the model group, the score of NAS decreased significantly, and the liver function and blood glucose were significantly improved. The level of serum CHOL, TG and LDL-C decreased to different degrees and HDL-C increased significantly at the same time. CONCLUSION: Fangzhengqi can improve liver histopathological changes and significantly reduce NAFLD activity score (NAS), which may be one of the mechanisms of its treatment of non-alcoholic steatohepatitis induced by high-fat and high-sugar diet.
