目的 研究As2 O3 对白血病细胞株NB4 ,K5 6 2 ,HL 6 0的活性氧自由基 (ROS)的影响。方法 细胞内活性氧 (ROS)用DCFH DA标记 ,并用流式细胞仪检测。结果 0 .6 μmol/LAs2 O3 作用 72h能诱导NB4发生 4 4 .9%的凋亡 ,提高其浓度至 2 .7和 8.1μmol/L ,也能诱导K5 6 2和HL 6 0分别发生 5 8.3%和 6 2 .3%凋亡。As2 O3 能显著降低三种细胞内ROS水平 ,且随浓度增加而降低。As2 O3 无论高浓度短时间作用或低浓度长时间作用对细胞内ROS抑制作用是相似的。结论 ROS水平下降在As2 O3 的诱导凋亡作用过程起了重要作用。
Objective To investigate the effect of As2 O3 on reactive oxygen species (ROS) in leukemia cell lines NB4, K5 6 2 and HL 60. Methods Intracellular reactive oxygen species (ROS) were labeled with DCFH DA and detected by flow cytometry. Results After treated with 0 .6 μmol / L As2 O3 for 72 h, 4 4 .9% apoptosis was induced in NB4 cells. When the concentration of As2 O3 was increased to 2.7 and 8.1 μmol / L, K5 6 2 and HL 6 0 5 8 8.3 % And 62.3% apoptosis. As2 O3 can significantly reduce the level of ROS in three kinds of cells, and decrease with increasing concentration. As2 O3 is similar to intracellular ROS inhibition regardless of high or short duration of action. Conclusion The decrease of ROS level plays an important role in the process of apoptosis induced by As2 O3.
