目的: 研究蓼科植物阴阳莲提取物 3, 4, 5-三羟基芪- 3-β-单- D-葡萄糖苷( 3, 4, 5,- trihydroxystibene- 3-β- mono- D- glucoside, THMG)对大肠癌细胞的体外生长特性、粘附性及浸润力的影响;从细胞层次探讨 THMG抗转移作用的基本环节。方法:检测 4种大肠癌细胞( HR8348, Hce8693, HT29, LoVo)在 THMG 0.4 mmol/L, 0.8 mmol/L, 1.6 mmol/L, 3.2 mmol/L等浓度作用下的存活率、生长曲线、粘附率及浸润力。结果:在实验浓度下, THMG对 4种大肠癌细胞的存活率、生长曲线无显著影响( P >0.05),在 3.2 mmol/L浓度时对高转移性的 LoVo细胞的粘附抑制率为 68.09% ,浸润抑制率为 89.95% ,对低转移性的 HT29细胞粘附抑制率为 78.75%、浸润抑制率为 93.31%,且随着浓度加大,粘附和浸润抑制率提高。结论: THMG可能通过抑制癌细胞粘附性和浸润力,发挥抗转移效应。
Objective: To study 3, 4, 5-trihydroxystilbene-3-β-mono-D-glucoside (3, 4, 5,-trihydroxystibene-3-β- mono-D- The effect of glucoside, THMG) on the in vitro growth characteristics, adhesion and infiltration of colorectal cancer cells; the basic aspects of anti-metastasis effect of THMG from the cell level. METHODS: Survival, growth curves, and viscidity of four colorectal cancer cells (HR8348, Hce8693, HT29, LoVo) at concentrations of 0.4 mmol/L, 0.8 mmol/L, 1.6 mmol/L, and 3.2 mmol/L of THMG were measured. With rate and infiltration. RESULTS: Under the experimental concentration, THMG had no significant effect on the survival rate and growth curve of the 4 colorectal cancer cells (P > 0.05). The adhesion inhibition rate of high metastatic LoVo cells was 68.09 at 3.2 mmol/L. %, the infiltration inhibition rate was 89.95%, the inhibition rate of adhesion to the low metastatic HT29 cells was 78.75%, and the infiltration inhibition rate was 93.31%. With the increase of the concentration, the inhibition rate of adhesion and infiltration increased. Conclusion: THMG may exert its anti-metastatic effect by inhibiting the adhesion and infiltration of cancer cells.
