目的观察加味四逆散(JWSND)对慢性轻度不可预计性心理应激大鼠海马损伤的影响。方法成年大鼠随机分为6组:雌性正常对照组,雌性模型组,雌性JWSND组,雄性正常对照组,雄性模型组,雄性JWSND组。建立慢性轻度不可预计性心理应激大鼠模型,比较造模前后糖水偏爱度;采用放射免疫方法检测大鼠下丘脑促肾上腺皮质激素释放激素(CRH)、血清促肾上腺皮质激素(ACTH)含量,2,3,5-氯化三苯基四氮唑(TTC)染色比色法测定大鼠海马脑片光密度(OD)值,流式细胞仪法检测海马神经元凋亡率。结果慢性心理应激大鼠糖水偏爱度明显下降(P<0.01或P<0.05),下丘脑-垂体轴功能亢进,海马脑片OD值明显低于正常对照组(P<0.01),细胞凋亡率明显升高(P<0.05或P<0.01);JWSND能明显提高心理应激大鼠糖水偏爱度(P<0.01),明显提高心理应激大鼠海马脑片OD值(P<0.05或P<0.01),降低海马神经元凋亡率(P<0.01)。结论 JWSND可有效减轻海马神经元损伤,此作用可能是其抗应激损伤中枢作用的主要机制之一。
Objective To observe the effects of JWSND on the hippocampal lesion in rats with chronic mild unpredictable psychological stress. Methods Adult rats were randomly divided into 6 groups: female normal control group, female model group, female JWSND group, male normal control group, male model group and male JWSND group. To establish a rat model of chronic mild unpredictable psychological stress, and to compare the degree of predilection for sugars and water before and after modeling. The levels of corticotropin-releasing hormone (CRH) and adrenocorticotropic hormone (ACTH) in the hypothalamus were measured by radioimmunoassay The optical density (OD) of rat hippocampal slices was measured by TTC staining and the apoptosis rate of hippocampal neurons was detected by flow cytometry. Results Chronic psychological stress significantly decreased the predilection of sugary water (P <0.01 or P <0.05), hypothalamic-pituitary axis hyperactivity, and OD value of hippocampal slices was significantly lower than that of normal control group (P <0.01) (P <0.05 or P <0.01). JWSND could significantly improve the degree of glycohydramnios (P <0.01), and significantly increase the OD of hippocampal slices in psychological stress rats (P <0.05 or P <0.05 or P <0.01) <0.01), and decreased the apoptosis rate of hippocampal neurons (P <0.01). Conclusion JWSND can effectively reduce the damage of neurons in the hippocampus, and this effect may be one of the main mechanisms of its anti-stress injury in the central nervous system.
