目的探讨胃黏膜病变中幽门螺杆菌(Hp)感染与环氧化酶-2(COX-2)、核转录因子(NF-κB)表达的关系。方法应用免疫组织化学染色方法(S-P法)分别检测COX-2、NF-κB蛋白在人慢性浅表性胃炎、慢性萎缩性胃炎、不典型增生及胃癌组织中的表达情况。应用Giemsa染色方法及快速尿素酶试验判定Hp感染。结果慢性萎缩性胃炎、不典型增生、胃癌各组Hp阳性者COX-2及NF-κB的阳性表达显著高于Hp阴性者,Hp阳性各组间COX-2和NF-κB的阳性率呈逐渐升高趋势。Hp阴性各组间COX-2和的NF-κB表达无显著性差异。COX-2与NF-κB在胃黏膜病变中的表达具有显著相关性。结论在胃黏膜病变中COX-2与NF-κB的表达与Hp感染有关,介导COX-2与NF-κB表达可能是Hp感染致癌的重要机制之一。
Objective To investigate the relationship between Helicobacter pylori (Hp) infection and cyclooxygenase-2 (COX-2) and nuclear factor-kappa B (NF-κB) expression in gastric mucosal lesions. Methods The expressions of COX-2 and NF-κB protein in human chronic superficial gastritis, chronic atrophic gastritis, dysplasia and gastric cancer were detected by immunohistochemical staining (S-P method). Hp infection was determined by Giemsa staining and rapid urease test. Results The positive expression of COX-2 and NF-κB in Hp positive patients with chronic atrophic gastritis, atypical hyperplasia and gastric cancer were significantly higher than those with negative Hp, the positive rates of COX-2 and NF-κB in Hp positive group were gradually Increase trend. There was no significant difference in the expression of COX-2 and NF-κB between Hp negative groups. COX-2 and NF-κB in gastric mucosal lesions have a significant correlation. Conclusions The expression of COX-2 and NF-κB in gastric mucosal lesions is related to Hp infection. Mediating the expression of COX-2 and NF-κB may be one of the important mechanisms of Hp infection.
