用聚丙烯酰胺凝胶等电聚焦法对 2 0个香菇原种菌丝体的酯酶和苹果酸脱氢酶同工酶进行了分析 ,结果在 2 0个被试原种中共检测出 37种酯酶同工酶和 2 0多种苹果酸脱氢酶同工酶 ,每一对原种的酯酶酶谱都有差异 ,按照典型的酶带分布型可以把苹果酸脱氢酶酶谱分成 5种类型 ;尽管观察到不同条件下培养物的个别同工酶的相对活力有变化 ,但每一菌株的每一同工酶酶带的等电点是恒定不变的。所以 ,用等电聚焦法分析酯酶和苹果酸脱氢酶同工酶的分布 ,可以用于辨别不同的香菇原种
Esterase and malate dehydrogenase isozymes of 20 mycorrhizal mushroom mycelia were analyzed by polyacrylamide gel isoelectric focusing. As a result, 37 isolates were detected in 20 tested progenies Esterase isozyme and 20 kinds of malate dehydrogenase isoenzyme, each pair of esterase isozyme spectrum are different, according to the typical distribution of the enzyme band malate dehydrogenase zymogram can be divided into 5 types; the isoelectric point of each isoenzyme enzyme band of each strain is constant, although changes in the relative activities of individual isoenzymes of the culture under different conditions are observed. Therefore, isoelectric focusing analysis of esterase and malate dehydrogenase isozymes distribution, can be used to identify different mushrooms
