目的探讨金黄色葡萄球菌(SAU)对人脐静脉内皮细胞(HUVEC)的黏附、内化及诱导细胞凋亡的发生、发展的机制和致病意义。方法常规方法制备SAU悬液,分别感染HUVEC细胞30 min、1、24、h。采用光学显微镜和荧光显微镜观察SAU对HUVEC的黏附、内化情况,采用流式细胞仪、DAPI荧光染色及DNA琼脂糖凝胶电泳分析SAU对HUVEC细胞诱导凋亡的影响。结果SAU与HUVEC共育30 min发现有细菌黏附细胞表面,1 h后有少量细菌侵入胞内,并伴有凋亡现象的发生,随着时间的延长,内化的细菌数没有增加,凋亡现象通过流式细胞仪、DAPI荧光染色及DNA琼脂糖凝胶电泳分析明显的被观测到。结论SAU通过黏附、内化侵入内皮细胞并诱导内皮细胞发生凋亡。
Objective To investigate the adhesion and internalization of Staphylococcus aureus (SAU) to human umbilical vein endothelial cells (HUVECs) and the mechanism and pathogenic significance of the apoptosis. Methods The SAU suspension was prepared by routine method and infected with HUVECs for 30 min, 1, 24, h respectively. The adhesion and internalization of SAU to HUVEC were observed with optical microscope and fluorescence microscope. The effect of SAU on apoptosis induced by HUVECs was analyzed by flow cytometry, DAPI fluorescence staining and DNA agarose gel electrophoresis. Results After 30 min of incubation with SAU and HUVEC, bacterial adhesion cells were found. After 1 h, a small amount of bacteria invaded the cells and accompanied with the appearance of apoptosis. With the extension of time, the number of internalized bacteria did not increase and apoptosis Phenomena were clearly observed by flow cytometry, DAPI fluorescence staining and DNA agarose gel electrophoresis. Conclusion SAU induces endothelial cell apoptosis through adhesion and internalization.
