目的 :研究褪黑素对人胃癌细胞SGC-7901黏附和迁移能力的影响及探索可能的分子机制。方法 :用不同浓度的褪黑素孵育人胃癌细胞SGC-7901 24 h,通过免疫印迹方法检测F-actin和G-actin蛋白表达水平,通过免疫荧光法检测细胞骨架结构的改变,通过细胞黏附实验观察胃癌细胞对血管内皮细胞黏附能力的变化,通过划痕实验检测褪黑素对细胞迁移能力的影响。结果:褪黑素使人胃癌细胞SGC-7901 F-actin量明显降低,同时细胞骨架结构发生重组,应力纤维出现断裂;褪黑素可抑制胃癌细胞的迁移及对血管内皮细胞的黏附能力。结论:褪黑素可能通过破坏细胞内应力纤维结构的稳定,最终导致了胃癌细胞的黏附和迁移能力的降低。
Objective: To investigate the effect of melatonin on the adhesion and migration of human gastric cancer cell line SGC-7901 and explore the possible molecular mechanism. Methods: Human gastric cancer cell line SGC-7901 was incubated with different concentrations of melatonin for 24 hours. The expression of F-actin and G-actin protein was detected by Western blotting. The cytoskeletal structure was detected by immunofluorescence. Cell adhesion assay The changes of adhesion ability of gastric cancer cells to vascular endothelial cells were observed. The influence of melatonin on the migration ability of cells was detected by scratch test. Results: Melatonin significantly decreased the amount of F-actin in human gastric cancer SGC-7901 cells, and resulted in the remodeling of cytoskeleton and stress fiber fracture. Melatonin inhibited gastric cancer cell migration and adhesion to vascular endothelial cells. CONCLUSION: Melatonin may reduce the adhesion and migration of gastric cancer cells by disrupting the stability of intracellular stress fibers.
