目的:观察川芎嗪对野百合碱(MCT)引起的肺动脉高压大鼠肺动脉压力及肺血管重构的影响。方法:将30只雄性SD大鼠随机分为3组各10只。MCT组和治疗组建立大鼠肺动脉高压模型。对照组、MCT组以生理盐水灌胃,每天2 mL,每天1次,连续20天;治疗组以川芎嗪80 mg/(kg·d),每天灌胃1次,连续20天。检测平均肺动脉压力(mPAP)、右心室肥大指数(RVHI)、肺血管增殖细胞核抗原(PCNA)和α-平滑肌肌动蛋(α-SMA)的表达。结果:对照组大鼠的mPAP低于MCT组(P<0.01);治疗组、对照组大鼠的mPAP、RVHI较MCT组降低(P<0.01);MCT组肺血管PCNA及α-SMA表达较治疗组、对照组增加。结论:川芎嗪可降低MCT引起的肺动脉高压大鼠肺动脉压力,减轻大鼠肺动脉重构。
AIM: To observe the effects of ligustrazine on pulmonary artery pressure and pulmonary vascular remodeling induced by monocrotaline (MCT) in rats with pulmonary hypertension. Methods: Thirty male Sprague-Dawley rats were randomly divided into three groups of 10 rats. The model of pulmonary hypertension was established in MCT group and treatment group. The control group and the MCT group were intragastrically administered with normal saline 2 mL daily for 20 consecutive days. The rats in the treatment group were treated with ligustrazine 80 mg / (kg · d) once daily for 20 consecutive days. The mean pulmonary arterial pressure (mPAP), RVHI, PCNA and α-SMA were measured. Results: The mPAP in the control group was lower than that in the MCT group (P <0.01). The mPAP and RVHI in the treatment group and the control group were lower than those in the MCT group (P <0.01) The treatment group and the control group increased. CONCLUSION: Ligustrazine can reduce pulmonary arterial pressure induced by MCT in rats with pulmonary hypertension and reduce pulmonary remodeling in rats.
