根据正交设计法,混合水平L32(8×4~2)设计,探讨小鼠骨髓细胞分裂的昼夜周期及其染色体制片最佳条件。实验设计内容为:每3小时杀一组动物,24小时共8次,即3、6、9、12、15、18、21、24点;秋水仙素剂量分3、4、5、6 mg/kg,其作用时间分别为2、3、4、6小时。实验结果表明,小鼠骨髓细胞的分裂有一定的昼夜周期和高峰,时间在上午9~10点和晚上9~12点(也可能是晚上9~10点),最低值在下午6点。
According to the orthogonal design method and mixed level L32 (8×4~2) design, the circadian cycle and the optimal conditions for chromosome production of mouse bone marrow cell division were explored. The experimental design includes: killing one group of animals every 3 hours, 8 times in 24 hours, ie 3, 6, 9, 12, 15, 18, 21, 24 points; 3, 4, 5, 6 mg of colchicine dose /kg, its effect time is 2, 3, 4, and 6 hours, respectively.
