目的通过构建寨卡病毒基因进化树以及比较氨基酸序列差异,了解寨卡病毒进化及基因变异情况。方法从GenBank获取1947年后各来源、各地区的寨卡病毒全基因组序列以及氨基酸序列,构建进化树并对氨基酸序列进行比较与分析。结果寨卡病毒非洲型与亚洲型毒株可各自聚类,2007年之后分离的毒株均为亚洲型;2016年分离自中国的14株寨卡病毒均为亚洲型,同源性高达99.1%~100.0%,氨基酸变异位点11个。结论寨卡病毒的分子进化与地理分布有密切的关系;2015-2016年分离的毒株同源性较高,序列较为保守;中国的14株毒株序列相近,其中1118、1875、2445以及2634 4个氨基酸位点可能对各自蛋白的结构和功能有较大的影响。
Objective To understand the evolution and gene mutation of Zika virus by constructing the evolutionary tree of Zika virus gene and comparing the amino acid sequence differences. Methods Genomic sequences and amino acid sequences of Zika virus were obtained from GenBank from all sources and regions. Phylogenetic tree was constructed and the amino acid sequences were compared and analyzed. Results The Zika virus strains of Africa and Asia could be clustered respectively. The strains isolated after 2007 were all Asian. The 14 Zika virus isolates from China in 2016 were all Asian and the homology was as high as 99.1% ~ 100.0%, 11 amino acid variation sites. Conclusion The molecular evolution of Zika virus is closely related to its geographical distribution. The homology of the strains isolated from 2015 to 2016 is high and the sequences are relatively conservative. The sequences of 14 strains in China are similar, with 1118, 1875, 2445 and 2634 Four amino acid sites may have a greater impact on the structure and function of each protein.
