目的 观察大鼠坐骨神经挤压伤后纤溶成分中组织型纤溶酶原激活剂 (t PA)和纤溶酶原激活剂抑制因子 1(PAI 1)蛋白表达的变化 ,了解t PA、PAI 1在周围神经损伤后修复过程中的作用。方法 取 2 1只Wistar大鼠 ,分别造成坐骨神经挤压伤模型。在术后 6h、1d、3d、7d、14d和 2 1d分别取材进行组织学、免疫组化观察。结果 术后 3d时神经断端回缩球处的t PA抗原呈阳性反应。 7d时损伤处的雪旺细胞明显增多 ,且t PA、PAI 1均有表达。 14d、2 1d时t PA表达有所减少 ,而PAI 1的表达则逐渐增多。结论 t PA和PAI 1参与周围神经挤压伤后的病理生理改变 ,在损伤神经修复过程中具有一定的作用。
Objective To observe the changes of the expression of tissue plasminogen activator (t PA) and plasminogen activator inhibitor 1 (PAI 1) in rats after sciatic nerve crush injury and find out the relationship between t PA and PAI 1 Role in the repair process after peripheral nerve injury. Methods Twenty-one Wistar rats were randomly divided into two groups: sciatic nerve crush injury model. At 6h, 1d, 3d, 7d, 14d and 21d after operation, the specimens were harvested for histological and immunohistochemical observation. Results The t PA antigen at the retracted bulb of the nerve was positive at 3d postoperatively. At 7 days, the number of Schwann cells in the lesion increased significantly, and both t PA and PAI 1 were expressed. At 14d and 21d, t PA expression decreased while PAI 1 expression gradually increased. Conclusion t PA and PAI 1 participate in pathophysiological changes after crush injury of peripheral nerves and play a certain role in the process of nerve repair.
