目的探讨肝癌衍生生长因子(Hepatoma-derived growth factor,HDGF)在乳腺癌组织中的表达及其意义。方法应用逆转录聚合酶链反应(Reverse-transcription polymerase chain reaction,RT-PCR)方法检测24例乳腺癌及其配对正常乳腺组织中HDGF mRNA的表达;应用免疫组织化学SP法检测75例乳腺癌及其配对正常乳腺组织中HDGF蛋白的表达水平。结果乳腺癌组织和正常乳腺组织中HDGF基因的表达强度分别是看家基因β-actin的(2.00±0.85)和(0.67±0.37)倍,正常乳腺组织中HDGF mRNA表达明显低于乳腺癌组织中的表达(P<0.001);HDGF蛋白在乳腺癌组织的表达水平明显高于正常乳腺组织(P<0.001);乳腺癌临床分期越高,HDGF的表达水平越高;淋巴结转移阳性的乳腺癌病例中HDGF的表达水平明显高于淋巴结转移阴性组,差异具有统计学意义(P>0.05)。结论 HDGF高表达可能促进了乳腺癌的发病过程,并与乳腺癌淋巴结转移相关。
Objective To investigate the expression of Hepatoma-derived growth factor (HDGF) in breast cancer and its significance. Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of HDGF mRNA in 24 cases of breast cancer and matched normal breast tissues. Immunohistochemical SP method was used to detect 75 cases of breast cancer and It paired with HDGF protein expression in normal breast tissue. Results The expression intensity of HDGF gene in breast cancer tissues and normal breast tissues were (2.00 ± 0.85) and (0.67 ± 0.37) times that of housekeeping genes, respectively. The expression of HDGF mRNA in normal breast tissues was significantly lower than that in breast cancer tissues (P <0.001). The expression of HDGF protein in breast cancer tissues was significantly higher than that in normal breast tissues (P <0.001). The higher the clinical stage of breast cancer, the higher the expression level of HDGF. The incidence of lymph node metastasis in breast cancer patients The expression of HDGF was significantly higher than that of lymph node metastasis negative group (P> 0.05). Conclusion High expression of HDGF may promote the pathogenesis of breast cancer and is associated with lymph node metastasis of breast cancer.
