目的观察泡球蚴感染BALB/c小鼠肝脏转化生长因子β1(TGF-β1)的动态变化。方法将60只BALB/c小鼠随机分为实验组和对照组,肉眼直视下肝脏穿刺感染泡球蚴。分别于感染后2 d8、d、4周、12周、24周、36周采取小鼠肝脏组织,通过HE和Masson染色观察小鼠肝脏病理改变,免疫组织化学法观察泡球蚴感染不同阶段的TGF-β1动态变化。结果泡球蚴感染小鼠肝脏汇管周围出现炎细胞浸润、脂肪变性、坏死、钙化和肝泡球蚴纤维囊壁形成等多种病理改变;TGF-β1表达水平呈先升高再下降趋势,感染4周、12周、24周、36周时的阳性率分别为(1.44±3.22)%、(18.83±4.03)%(、9.44±4.71)%和(8.13±3.65)%,其中感染12周和36周与对照组相比差异有统计学意义(P<0.05)。结论泡球蚴感染BALB/c小鼠TGF-β1表达上调,可能与感染小鼠肝纤维化的发展有关。
Objective To observe the dynamic changes of hepatic transforming growth factor β1 (TGF-β1) in BALB / c mice infected by cysticercus. Methods Sixty BALB / c mice were randomly divided into experimental group and control group. The liver puncture infected the cysticercus with naked eyes. The liver tissues of mice were taken at 2 d 8, d, 4, 12, 24 and 36 weeks after infection respectively. Pathological changes of liver were observed by HE and Masson staining. Immunohistochemistry was used to observe the different stages of infection of metacercariae TGF-β1 dynamic changes. Results A number of pathological changes such as inflammatory cell infiltration, steatosis, necrosis, calcification and cystic fibrosis of hepatic hydatid cyst were found in the periportal tract of mice infected with cysticercus. The expression of TGF-β1 increased first and then decreased, The positive rates were (1.44 ± 3.22)%, (18.83 ± 4.03)% (9.44 ± 4.71)% and (8.13 ± 3.65)% at 4 weeks, 12 weeks, 24 weeks and 36 weeks respectively. And 36 weeks compared with the control group, the difference was statistically significant (P <0.05). Conclusion The up-regulation of TGF-β1 in BALB / c mice infected by cysticercosis may be related to the development of hepatic fibrosis in mice infected with BALB / c mice.
