目的研究尼妥珠单抗(h-R3,泰欣生)联合γ射线照射对人食管癌Ec109细胞生长的抑制作用。方法利用四氮唑盐(MTT)比色法分析h-R3、照射及两者联合对人食管癌Ec109细胞的生长抑制作用,通过流式细胞仪分析细胞周期分布及细胞凋亡的变化。结果 h-R3组、照射组和联合照射组对Ec109细胞生长均有抑制作用,且h-R3对Ec109细胞生长抑制作用呈时间和剂量依赖性,联合照射组对Ec109细胞的生长抑制作用最强(45.77%),明显高于h-R3组(31.48%)和照射组(35.71%)(F=21.08,P<0.05)。联合照射组细胞出现明显的G2期阻滞和细胞凋亡,G2期细胞和凋亡细胞所占比例最高,分别达到31.67%和23.62%。结论尼妥珠单抗体外有效抑制人食管癌Ec109细胞的生长,与γ射线照射联合后抑瘤作用具有一定程度的协同效应。
Objective To study the inhibitory effect of nimotuzumab (h-R3, Taixin Health) combined with γ-ray irradiation on the growth of human esophageal cancer Ec109 cells. Methods The growth inhibition of human esophageal carcinoma Ec109 cells treated with h-R3, irradiation and their combination was analyzed by MTT assay. The cell cycle distribution and apoptosis were analyzed by flow cytometry. Results The growth of Ec109 cells was inhibited in h-R3 group, irradiation group and combined irradiation group, and the growth inhibitory effect of h-R3 on Ec109 cells was time-and dose-dependent. The combined irradiation group had the strongest inhibitory effect on the growth of Ec109 cells (45.77%), which was significantly higher than that of h-R3 group (31.48%) and irradiation group (35.71%) (F = 21.08, P <0.05). The combination of irradiation group G2 cells showed obvious G2 arrest and apoptosis, G2 phase cells and apoptotic cells accounted for the highest proportion, respectively 31.67% and 23.62%. Conclusion Nimotuzumab can effectively inhibit the growth of human esophageal cancer Ec109 cells in vitro, and it has some synergistic effects after combined with γ-ray irradiation.
