目的检测卵巢上皮癌中缺氧诱导因子-1α(HIF-1α)、葡萄糖转运蛋白-1(GLUT-1)蛋白的表达,探讨二者的相关性及临床意义。方法免疫组化法检测35例卵巢上皮性癌、15例卵巢交界性肿瘤、20例卵巢良性肿瘤及20例正常卵巢组织中HIF-1α、GLUT-1蛋白的表达。结果相对于卵巢良性肿瘤和正常卵巢组织,HIF-1α、GLUT-1蛋白在卵巢上皮癌和交界性肿瘤中表达明显升高,差异有统计学意义(P<0.05),而在正常卵巢组织及卵巢良性肿瘤中不表达或低表达。卵巢上皮癌中,HIF-1α蛋白表达水平与患者年龄、肿瘤的病理类型以及肿瘤的分化程度无明显相关性,差异无统计学意义(P>0.05),而与肿瘤的临床分期相关,差异有统计学意义(P<0.05);GLUT-1蛋白表达水平与肿瘤的分化程度及临床分期有明显的相关性,差异有统计学意义(P<0.05),而与患者年龄及肿瘤的病理类型无明显相关性,差异无统计学意义(P>0.05)。HIF-1α与GLUT-1蛋白的表达呈正相关(r=0.623,P<0.01)。结论在卵巢上皮癌中,HIF-1α、GLUT-1蛋白的表达升高可能促进了卵巢上皮癌的发生、发展;HIF-1α蛋白作为一种核转录因子,可能调控了GLUT-1蛋白的表达。HIF-1α、GLUT-1蛋白有望成为卵巢上皮癌的新的肿瘤标记物及治疗的靶点。
Objective To detect the expression of hypoxia inducible factor-1α (HIF-1α) and glucose transporter protein-1 (GLUT-1) protein in ovarian epithelial carcinoma and to explore their correlation and clinical significance. Methods The expressions of HIF-1α and GLUT-1 protein in 35 cases of ovarian epithelial carcinoma, 15 cases of ovarian borderline tumor, 20 cases of benign ovarian tumor and 20 cases of normal ovarian tissue were detected by immunohistochemistry. Results Compared with benign ovarian tumor and normal ovarian tissue, the expression of HIF-1α and GLUT-1 protein in epithelial ovarian tumor and borderline tumor was significantly increased (P <0.05), while in normal ovarian tissue and Ovarian benign tumor is not expressed or low expression. In epithelial ovarian cancer, the expression level of HIF-1αwas not related to the patient’s age, the pathological type of the tumor and the degree of tumor differentiation (P> 0.05), but was related to the clinical stage of the tumor (P <0.05). There was a significant correlation between the expression of GLUT-1 protein and tumor differentiation degree and clinical stage (P <0.05), but not with age and tumor pathological type Significant correlation, the difference was not statistically significant (P> 0.05). HIF-1α and GLUT-1 protein expression was positively correlated (r = 0.623, P <0.01). Conclusion The increased expression of HIF-1α and GLUT-1 may promote the occurrence and development of epithelial ovarian cancer in ovarian epithelial carcinoma. HIF-1α protein may regulate the expression of GLUT-1 protein as a nuclear transcription factor . HIF-1α, GLUT-1 protein is expected to become a new tumor marker for ovarian epithelial cancer and the treatment of the target.
