目的分离和测定Naftopidil手性代谢产物:去甲基代谢产物(DMN)、苯羟化代谢产物(PHN)和萘羟化代谢产物(NHN)的对映体。方法采用HPLC法,流动相为异丙醇-正己烷(15∶85),流速为1.0 mL.min-1,柱温为25℃,检测波长为283 nm。结果 3对对映体能同时得到良好地分离。经方法学验证,R-DMN(4.99~159.68μg.mL-1)、R-PHN(5.30~168.00μg.mL-1)、R-NHN(5.00~160.16μg.mL-1)与峰面积线性良好;RSD<1.03%。结论所用方法简单、快速、准确,可同时测定合成的R-DMN、R-PHN和R-NHN的含量,为Naftopidil手性药物代谢学研究提供了方法。
OBJECTIVE To separate and determine the enantiomers of Naftopidil chiral metabolites: demethylated metabolites (DMNs), benzylated hydroxylated metabolites (PHNs), and Naphthylated metabolites (NHNs). Methods The HPLC method was used. The mobile phase was isopropanol-n-hexane (15:85) at a flow rate of 1.0 mL · min-1. The column temperature was 25 ℃ and the detection wavelength was 283 nm. Results Three enantiomers were well separated at the same time. R-DMN (4.99-159.68μg.mL-1), R-PHN (5.30-168.00μg.mL-1) and R-NHN (5.00-160.16μg.mL-1) Good; RSD <1.03%. Conclusion The method is simple, rapid and accurate, and the contents of R-DMN, R-PHN and R-NHN can be determined simultaneously. This provides a method for the study of Naftopidil chiral drug metabolism.
