目的研究p,p’-DDE对离体培养的大鼠睾丸支持细胞细胞内抑制素B的影响。方法从大鼠睾丸组织中分离支持细胞进行离体原代培养,在细胞毒性试验的基础上确定受试物的剂量,以RT-PCR方法定量抑制素B在不同剂量受试物作用下的变化情况。结果p,p’-DDE中、高浓度组与溶剂对照组和低浓度组比较差异均有显著性(P<0.05)。抑制素B的灰度值随着p,p’-DDE浓度的增高逐渐降低,呈剂量依赖关系。结论p,p’-DDE对大鼠睾丸支持细胞分泌抑制素B表达可能存在着抑制作用。
Objective To investigate the effect of p, p’-DDE on endostatin B in cultured rat sertoli cells. Methods Sertoli cells were isolated from rat testis for primary culture in vitro. The dose of test substance was determined on the basis of cytotoxicity test. The changes of inhibin B under different doses of test substance were determined by RT-PCR Happening. Results There was significant difference between p and p’-DDE in high concentration group and solvent control group and low concentration group (P <0.05). The gray value of inhibin B decreased with the increase of p, p’-DDE concentration in a dose-dependent manner. Conclusion p, p’-DDE may inhibit the expression of inhibin B in the sertoli cells of rats.
