目的分析1型糖尿病(T1DM)患者外周血浆细胞样树突状细胞(PDC)数量及功能变化。方法采用流式细胞术检测22例T1DM患者(DM组)及20位健康人(NC组)体内PDC的比例及数量;用免疫刺激序列(CpG ODN-2216)刺激外周血单个核细胞(PBMCs)并与之共培养,通过检测PBMCs产生的干扰素α(IFN-α)水平评估PDC的功能。结果与NC组比较,DM组外周血PDC的百分比和数量均显著下降(P<0.01);PDC经CpG ODN-2216刺激后产生IFN-α的能力也明显下降(P<0.05);相关性分析表明,经CpG ODN-2216刺激后IFN-α的产量与PDC数量呈正相关(r=0.439,P<0.05)。结论 T1DM患者体内PDC数量和功能异常,说明PDC可能参与了机体免疫功能紊乱进程;PDC产生的IFN-α减少,可能使患者对感染的控制能力降低而易感。
Objective To analyze the changes of peripheral plasmacytoplasmic dendritic cells (PDC) in patients with type 1 diabetes mellitus (T1DM). Methods Flow cytometry was used to detect the proportion and number of PDC in 22 T1DM patients and 20 healthy subjects (NC group). PBMCs were stimulated with CpG ODN-2216, And co-cultured to assess the function of PDC by measuring the level of interferon alpha (IFN-a) produced by PBMCs. Results Compared with NC group, the percentage and the number of PDC in peripheral blood of DM group decreased significantly (P <0.01); the ability of PDC to produce IFN-α after CpG ODN-2216 stimulation decreased significantly (P <0.05); Correlation analysis The results showed that the production of IFN-α was positively correlated with the number of PDC after stimulation with CpG ODN-2216 (r = 0.439, P <0.05). Conclusion The number and dysfunction of PDC in T1DM patients indicate that PDC may participate in the process of immune dysfunction. The decrease of IFN-α produced by PDC may make the patients susceptible to the control of infection.
