目的 :研究新生隐球菌白化突变分子机制。 方法 :采用聚合酶链反应 (PCR)技术对新生隐球菌产黑素株与白化突变株进行比较研究 ,并进行 PCR产物的克隆 ,构建重组表达质粒 ,转化大肠杆菌。 结果 :发现白化突变株酚氧化酶结构基因 (CNL AC1)在 1715~ 2 6 17bp之间有大片段缺失 (约 45 0 bp) ,而产黑色素株却无此缺失片段 ,成功地进行了质粒载体的构建。 结论 :本研究为新生隐球菌白化突变分子机制的研究提供帮助 ,为新生隐球菌病的预后和基因治疗提供可能的靶基因。
Objective: To study the molecular mechanism of Cryptococcus neoformans albino mutation. Methods: Polymerase chain reaction (PCR) was used to compare the melanin-producing strains and albino mutant strains of Cryptococcus neoformans. The PCR products were cloned and the recombinant expression plasmids were constructed and transformed into Escherichia coli. Results: It was found that there was a large fragment deletion (about 45 0 bp) in the albinism mutant phenoloxidase gene (CNL AC1) between 1715 and 2617 bp, but no deletion fragment was found in the melanin producing strain. Conclusion: This study is helpful for studying the molecular mechanism of Cryptococcus neoformans albino mutation, providing possible target genes for the prognosis and gene therapy of cryptococcosis of newborn.
