目的:研究槲皮素纳米乳在大鼠体内各肠段的吸收情况,并考察其形态学及粒径分布。方法:采用大鼠在体小肠各段回流实验,紫外分光光度法测定药物浓度,根据药物在小肠各段中的减少量来确定药物的吸收;采用透射电子显微镜观察纳米乳形态。结果:槲皮素纳米乳在大鼠十二指肠、空肠、回肠及结肠中的吸收速率常数(ka)依次为0.0675,0.1056,0.1892,0.1615 h~(-1);透射电镜下槲皮素纳米乳呈球形,大小均匀,平均粒径(16.3±4.6)nm;结论:槲皮素纳米乳在小肠下段吸收较好,吸收呈一级动力学过程。
OBJECTIVE: To study the absorption of quercetin nanoemulsion in various intestinal segments of rats, and to investigate its morphology and particle size distribution. METHODS: Rats were used for reflux experiments in each segment of the small intestine. UV spectrophotometry was used to determine drug concentration. The drug absorption was determined according to the reduction of the drug in each segment of the small intestine. Transmission electron microscopy was used to observe the nanoemulsion morphology. RESULTS: The absorption rate constants (ka) of quercetin nanoemulsion in duodenum, jejunum, ileum and colon were 0.0675, 0.1056, 0.1892, 0.1615 h-1, respectively; quercetin was identified by transmission electron microscope. The nanoemulsion was spherical, uniform in size and average particle size (16.3±4.6) nm. Conclusion: Quercetin nanoemulsion absorbs well in the lower part of the small intestine, and the absorption takes a first-order kinetic process.
