目的建立分离提取高纯度卡那霉素B的最佳工艺条件。方法以基因工程黑暗链霉菌spu46发酵产物为原料,通过对树脂吸附性能的研究,筛选吸附容量最大的弱酸性树脂;考察上样液的pH、浓度、流速等因素对吸附效果的影响;考察不同洗脱条件对分离提纯效果的影响。结果弱酸性阳离子树脂CD-180在pH 7.5、流速0.85 mL.m in-1的条件下,吸附量达到最高。0.15 mol.L-1的氨水在1.00 mL.m in-1,的流速下,解析率稳定,可达到85%以上。结论弱酸性阳离子树脂CD-180对卡那霉素B的分离纯化效果最佳。
Objective To establish the optimum conditions for the isolation and purification of kanamycin B with high purity. Methods The objective of the study was to screen the weak acid resin with the highest adsorption capacity by studying the adsorption properties of the resin by using the genetically engineered dark chain spu46 fermentation product. The influence of pH, concentration and flow rate on the adsorption was investigated. Effect of Elution Conditions on Separation and Purification. Results Weakly acidic cation resin CD-180 reached the highest adsorption capacity at pH 7.5 with a flow rate of 0.85 mL.m in-1. Ammonia water of 0.15 mol·L-1 at a flow rate of 1.00 mL.m in-1, the resolution is stable, up to 85%. Conclusion Weak acid cation resin CD-180 has the best effect on the isolation and purification of kanamycin B.
