基于血浆代谢组学分析金欣口服液治疗小鼠RSV肺炎的作用机制

目的:通过GC-MS技术分析BALB/c小鼠血浆内源性代谢产物的相对含量变化,阐明金欣口服液治疗呼吸道合胞病毒(RSV)肺炎的作用机制。方法:RSV滴鼻感染BALB/c小鼠建立RSV肺炎模型,首次滴鼻2 h后予金欣口服液(剂量27.6 g·kg-1·d-1)灌胃治疗,连续滴鼻3 d后各组处死5只小鼠取肺组织行组织病理学检查,连续给药7 d后采集各组小鼠的血浆,运用GC-MS检测(m/z 50~500)血浆内源性代谢物及其相对含量的变化,分析其可能参与的代谢通路。结果:RSV肺炎小鼠的血浆中共测得32种内源性代谢产物,其中乳酸、尿素、谷氨酰胺、葡萄糖、花生四烯酸和1,5-脱水山梨醇为差异代谢产物,主要与花生四烯酸、谷氨酰胺、丙酮酸的代谢通路有关。结论:金欣口服液治疗RSV肺炎可能与调节RSV感染导致的代谢紊乱相关。 OBJECTIVE: To analyze the changes of plasma endogenous metabolites in BALB / c mice by GC-MS and elucidate the mechanism of Jinxin oral liquid in the treatment of respiratory syncytial virus (RSV) pneumonia. Methods: BALB / c mice were infected with RSV intranasally to establish RSV pneumonia model. After the first intranasal instillation for 2 hours, Jinxin oral liquid (27.6 g · kg-1 · d-1) Five mice in each group were sacrificed for histopathology examination. After seven days of continuous administration, the plasma of each group of mice was collected. The plasma endogenous metabolites were detected by GC-MS (m / z 50-500) Its relative content changes, analysis of its possible involvement of the metabolic pathway. Results: A total of 32 endogenous metabolites were detected in the plasma of RSV pneumonia mice. Among them, lactic acid, urea, glutamine, glucose, arachidonic acid and 1,5-sorbitol were the differential metabolites, Tetraenoic acid, glutamine, pyruvate metabolic pathway related. Conclusion: Jinxin Oral Liquid treatment of RSV pneumonia may be related to the regulation of metabolic disorders caused by RSV infection.
