目的探讨补肾活血颗粒对帕金森病模型大鼠黑质、纹状体中Fas、FADD表达的影响。方法 60只雄性SD大鼠随机分为正常组10只和造模组50只,采用6-羟基多巴胺注射造模。造模成功的23只随机分为模型组10只和观察组13只。观察组给予补肾活血颗粒灌胃,正常组和模型组给予等量生理盐水灌胃。应用免疫组化染色技术观察大鼠黑质、纹状体中Fas和Fas死亡结构域蛋白(FADD)表达的变化情况。结果模型组Fas在黑质阳性细胞表达率(PI)为(1.400±0.070),纹状体PI为(1.820±0.090),均高于正常组和观察组(P<0.01)。模型组FADD在黑质PI为(1.160±0.058),纹状体PI为(0.780±0.040),均高于正常组和观察组(P<0.01)。结论补肾活血颗粒治疗帕金森病的疗效机制可能与下调Fas和FADD表达有关。
Objective To investigate the effect of Bushen Huoxue Granule on the expression of Fas and FADD in the substantia nigra and striatum of Parkinson’s disease rats. Methods Sixty male Sprague-Dawley rats were randomly divided into normal group (n = 10) and model group (n = 50). The rats were injected with 6-hydroxydopamine. Twenty-three successful models were randomly divided into model group (n = 10) and observation group (n = 13). Observation group was given Bushen Huoxue granule gavage, normal group and model group given the same amount of saline gavage. The changes of Fas and Fas death domain protein (FADD) expression in the substantia nigra and striatum of rats were observed by immunohistochemical staining. Results The positive rate of Fas in substantia nigra was (1.400 ± 0.070) and that of striatum was (1.820 ± 0.090) in model group, both of which were higher than those in normal group and observation group (P <0.01). The PI of substantia nigra in model group was (1.160 ± 0.058) and that of striatum was (0.780 ± 0.040), both of which were higher than those of normal group and observation group (P <0.01). Conclusion The therapeutic mechanism of Bushen Huoxue Granule in the treatment of Parkinson’s disease may be related to the down-regulation of the expression of Fas and FADD.
