首次采用特殊方法从感染兔红细胞上分离、纯化两种血吸虫组分抗原SjR47和SjR12,实验结果证明,前者属卵源性抗原,后者为虫源性抗原.两次实验结果均表明,两种组分抗原能诱导出明显的抗病免疫力,实验1获得54.95%-72.36%的肝内减卵率,实验2获得52.57%-57.27%的肝内减卵率和50.73%-74.70%的成熟卵减少率,并均可使肝内肉芽肿病变明显减小,尤以SjR47效果显著.两种抗原诱导的抗肝内肉芽肿保护性免疫调节机制的研究结果表明,两者均能诱导小鼠产生高水平的IgG,测定IgG及其亚类动态显示,在感染后0-12wk时,免疫小鼠血清IgG和IgG_1水平均显著高于佐剂和感染对照组者(P<0.01),特别是能反映保护性免疫力指标的IgG_1升高非常明显:SjR47免疫鼠的脾T淋巴细胞亚群表型动态研究显示,SjR47免疫鼠的CD4~+和CD8~+T细胞在感染4wk时均升高,随后下降.CD4~+/CD8~+比值也随时间延长而下降,呈现免疫下调,其变化趋势同肉芽肿病变减小相一致;SjR47免疫鼠的细胞因子水平动态观察结果表明,SjR47免疫鼠后可使TNF-α在小鼠体内的活性水平较平显著上升(成虫期前),减弱了TNF-α对虫卵肉芽肿形成的介导作用以及对雌虫排卵的促进作用,导致肝内肉芽肿病变减小和虫卵减少。本研究结果表明,两
