目的:研究药用真菌粗毛黄褐孔菌的生物学特性,为粗毛黄褐孔菌人工培养和工业化生产培养提供参考。方法:考察粗毛黄褐孔菌菌株在不同温度、光照下对菌丝生长的影响,酯酶同工酶酶谱。同时对液体培养粗毛黄褐孔菌的碳源、氮源和初始pH对菌丝生物量和胞外多糖含量的影响进行了研究。结果:粗毛黄褐孔菌菌丝在25℃生长速度较快,黑暗培养有利于菌丝生长,粗毛黄褐孔菌有8条酯酶同工酶。在液体培养条件下,综合考虑生物量和胞外多糖产量,葡萄糖是较好的碳源,酵母膏是较好的氮源,在6.5~7.5范围内的初始pH值较好。结论:为粗毛黄褐孔菌人工培养和工业化液体深层发酵培养提供参考。
Objective: To study the biological characteristics of the medicinal fungi rough brown fungus, to provide a reference for artificial cultivation and industrial production culture of the rough brown fungus. Methods: The effects of different strains on the mycelium growth and the esterase isozymes were studied under different temperatures and light conditions. At the same time, the effects of carbon source, nitrogen source and initial pH on the mycelia biomass and extracellular polysaccharide content of liquid culture of T. gmelinii were studied. Results: The mycelial growth rate of T. gmelinii at 25 ℃ was faster than that of the control. The dark culture was conducive to mycelial growth. There were 8 esterase isozymes in T. gmelinii. Under liquid culture conditions, glucose is a good carbon source considering the biomass and yield of exopolysaccharide, yeast extract is a good nitrogen source, and the initial pH value in the range of 6.5-7.5 is better. Conclusion: It provides a reference for the artificial culture of T. gmelinii and industrial liquid deep fermentation.
