目的:探讨解毒化瘀方中药血清对白血病K562/A02耐药细胞耐药逆转作用及NF-κB信号表达的影响。方法:采用MTT法检测解毒化瘀方中药血清对K562/A02细胞毒作用,流式细胞仪检测解毒化瘀方中药血清对K562/A02细胞内化疗药物浓度的影响,用Western blot法检测解毒化瘀方中药血清对NF-κB/P65、NF-κB/P50、IκBα的表达的影响。结果:解毒化瘀方中药血清对K562/A02细胞生长抑制作用呈浓度依赖性,能够增加K562/A02细胞内化疗药物的浓度,下调K562/A02细胞NF-κB/P65、NF-κB/P50、IκBα表达,从而降低NF-κB信号。结论:解毒化瘀方中药血清能够逆转K562/A02细胞耐药,其耐药逆转机制可能与下调K562/A02细胞NF-κB信号表达有关。
Objective: To investigate the effect of Jiedu Huayu Recipe serum on drug-resistant reversal and NF-κB signaling in K562 / A02 cell line. Methods: MTT was used to detect the cytotoxic effect of Jiedu Huayu Recipe serum on K562 / A02 cells. Flow cytometry was used to detect the effect of Jiedu Huayu Recipe serum on chemotherapeutic drug concentration in K562 / A02 cells. The detoxification was detected by Western blot Effect of Chinese herbs for removing stasis on the expression of NF-κB / P65, NF-κB / P50 and IκBα. Results: The serum of K562 / A02 cells inhibited the growth of K562 / A02 cells in a concentration - dependent manner, and could increase the concentration of chemotherapeutic drugs in K562 / A02 cells and down - regulate the expressions of NF-κB / P65 and NF-κB / IκBα expression, thereby reducing NF-κB signaling. Conclusion: The drug serum of Jiedu Huayu Recipe can reverse the drug resistance of K562 / A02 cells, and its drug resistance reversal mechanism may be related to the down-regulation of NF-κB signal expression in K562 / A02 cells.
