目的:通过建立家兔颈总动脉球囊损伤动脉粥样硬化模型,观察益气活血中药和辛伐他汀西药对家兔血管内皮功能的影响,研究益气活血复方抗动脉粥样硬化的作用机制。方法:实验选取普通雄性新西兰大白兔38只,随机分成正常对照组(8只),辛伐他汀组;益气活血复方组;模型组(均10只)。适应性喂养1周后,后3组行颈总动脉球囊损伤术。术后正常组给予普通饲料,其余各组给予高脂饲料。各组分别于第0周、4周、8周空腹耳中央动脉采血测定血浆NO含量,内皮素-1水平,并取颈总动脉组织观察形态学变化。结果:球囊损伤后第4周末,模型组兔血浆NO含量较正常对照组明显降低(P<0.01),辛伐他汀组和益气活血复方组兔血浆NO含量较同期模型组明显升高(P<0.01),其中益气活血复方组兔血浆NO水平升高较辛伐他汀组更为明显(P<0.05)。球囊损伤后第4周末,模型组家兔血浆ET-1水平较正常对照组明显上升;与模型组比较,辛伐他汀组及益气活血复方组血浆ET-1水平均显著下降(P<0.05),但两组间无显著性差异(P>0.05)。辛伐他汀组及益气活血复方组颈总动脉组织形态学改变较同期模型组有明显改善。结论:益气活血复方可促进NO合成和分泌,抑制ET-1表达,有利于恢复NO/ET-1平衡,能明显改善动脉粥样硬化组织形态的变化,提示益气活血复方可能具有的内皮恢复及改善功能在防止AS发生中有积极作用。
OBJECTIVE: To establish an animal model of atherosclerosis induced by balloon injury of the common carotid artery in rabbits and observe the effect of Yiqi Huoxue Chinese medicine and simvastatin western medicine on the vascular endothelial function in rabbits. To study the mechanism of Yiqi Huoxue Compound in preventing atherosclerosis . Methods: Thirty-eight male New Zealand white rabbits were randomly divided into normal control group (n = 8), simvastatin group, Yiqi Huoxue Compound group and model group (n = 10). After 1 week of adaptive feeding, the carotid artery balloon injury was performed in the last 3 groups. Normal group after giving normal feed, and the remaining groups were given high-fat diet. Plasma concentrations of NO and ET-1 were measured at 0, 4, and 8 weeks in each group. Blood samples were collected from the central aortic artery of fasting ears. The morphology of the common carotid artery was observed. Results: At the end of the 4th week after balloon injury, the content of NO in plasma in model group was significantly lower than that in normal control group (P <0.01), while the NO content in simvastatin group and Yiqi Huoxue Compound group was significantly higher than that in the model group P <0.01). The level of plasma NO in Yiqi Huoxue Compound-treated group was higher than that in simvastatin group (P <0.05). At the end of the fourth week after balloon injury, the ET-1 level in the model group was significantly higher than that in the normal control group. Compared with the model group, ET-1 levels in the simvastatin group and Yiqi Huoxue Compound group were significantly decreased (P < 0.05), but no significant difference between the two groups (P> 0.05). Compared with the model group, the changes of carotid artery morphology of simvastatin group and Yiqi Huoxue Compound group were significantly improved. Conclusion: Yiqi Huoxue Compound can promote NO synthesis and secretion, inhibit the expression of ET-1, restores the balance of NO / ET-1, and can obviously improve the changes of atherosclerosis. It suggests that Yiqi Huoxue Compound may have endothelium Recovery and improvement have a positive effect in preventing the occurrence of AS.
