目的探讨hCG联合免疫球蛋白在免疫性流产的治疗中是否具有协同作用。方法 CBA/J雌性小鼠按体重编号后按随机数字表法随机分为4组,即自然流产模型组(CBA/J×DBA/2)、hCG用药组(CBA/J×DBA/2,hCG 35U/kg,肌注,隔天一次)、hCG联合免疫球蛋白组(CBA/J×DBA/2,hCG 35U/kg,肌注,隔天一次,免疫球蛋白,40mg/kg,肌注,孕第一天)。正常妊娠模型组(CAB/J×BALB/c)。孕第14天处死孕鼠,计算各组胚胎吸收率,留取蜕膜并进行组织匀浆,ELISA方法测定蜕膜匀浆液中TNF-α、IFN-γ、IL-4、IL-10的含量。结果自然流产组的胚胎吸收率高于其它三组,差异有统计学意义(P<0.05),hCG组的胚胎吸收率高于联合用药组,差异有统计学意义(P>0.05)。与正常妊娠组比较,自然流产模型组TNF-α、IFN-γ的水平明显高于前者(P<0.05),而IL-4及IL-10的水平低于前者,差异均有统计学意义(P<0.05)。hCG组和联合用药组TNF-α及IFN-γ水平明显低于自然流产组(P<0.05),而IL-4及IL-10高于自然流产组,联合用药组及正常妊娠组细胞因子水平差异无统计学意义(P<0.05)。结论 hCG联合免疫球蛋白通过调节母胎界面Th1/Th2型细胞因子水平来促进妊娠成功,二者具有协同作用。
Objective To investigate whether hCG combined immunoglobulin has a synergistic effect in the treatment of immune abortion. Methods Female CBA / J mice were divided into four groups randomly according to body weight: CBA / J × DBA / 2, CBA / J × DBA / 2, hCG Intramuscular injection, once every other day, immunoglobulin, 40 mg / kg, intramuscular injection, intramuscular injection, intramuscular injection, First day of pregnancy). Results The embryo absorption rate of spontaneous abortion group was higher than that of the other three groups (P <0.05). The embryo absorption rate of hCG group was higher than that of combination group (P> 0.05). The levels of TNF-α and IFN-γ in spontaneous abortion model group were significantly higher than those in normal pregnancy group (P <0.05), while the levels of IL-4 and IL-10 were lower than those in normal pregnancy group P <0.05). The levels of TNF-α and IFN-γ in hCG group and combination group were significantly lower than those in spontaneous abortion group (P <0.05), while the levels of IL-4 and IL-10 were higher than those in spontaneous abortion group, combination group and normal pregnancy group The difference was not statistically significant (P <0.05). Conclusions hCG combined immunoglobulin can promote the success of pregnancy by regulating the level of Th1 / Th2 cytokines in the maternal-fetal interface, and the two have synergistic effects.
