目的 探讨胰岛素样生长因子Ⅰ类受体 (IGF ⅠR)与脐血B淋巴细胞增殖以及IgM合成的关系。方法 通过反义核酸阻抑脐血B淋巴细胞IGF ⅠR基因表达水平 ,采用同位素掺入法观察B淋巴细胞的增殖功能 ,利用酶联免疫吸附 (ELISA)双抗夹心法检测IgM含量 ,通过逆转录聚合酶链反应技术检测IGF ⅠR基因表达。结果 经反义寡核苷酸 (ASON)阻抑后 ,脐血B淋巴细胞IGF ⅠRmRNA的表达水平明显降低 ,降低幅度波动于 15 %~ 4 9%;同时B淋巴细胞的增殖功能较单纯美洲商陆 (PWM)刺激下降 5 4 %;细胞活化第 5天IgM的生成量明显降低。结论 IGF ⅠR参与脐血B淋巴细胞增殖、分化以及IgM的产生过程 ,提示IGF ⅠR与IGF Ⅰ结合发挥的生物活性在维持新生儿时期体液免疫水平方面起着重要的促进作用。
Objective To investigate the relationship between insulin-like growth factor type Ⅰ receptor (IGF ⅠR) and the proliferation of cord blood B lymphocytes and the synthesis of IgM. Methods Antisense oligonucleotide was used to inhibit the expression of IGF Ⅰ R gene in cord blood B lymphocytes. The proliferation of B lymphocytes was observed by isotope incorporation method. The IgM level was detected by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) Polymerase chain reaction to detect IGF Ⅰ R gene expression. Results After ASON suppression, the expression of IGF Ⅰ RmRNA in cord blood B lymphocytes was significantly decreased by 15% -49%, while the proliferative function of B lymphocytes was lower than that of pure American businessmen Landslide (PWM) stimulation decreased by 54%; on the fifth day of cell activation, IgM production decreased significantly. Conclusion IGF ⅠR is involved in the proliferation, differentiation and IgM production of B lymphocytes in cord blood, suggesting that the biological activity of IGF ⅠR binding to IGF Ⅰ plays an important role in maintaining humoral immunity during neonatal period.
