目的进一步研究FTY720的降低外周血淋巴细胞的效果和剂量及用药时间的关系。方法给予FTY72010mg/(kg.d),于给药前、第1次给药后2、4、8、24、48h采血计数外周血淋巴细胞数目;给予不同剂量的FTY720(0.3、1、3、10mg/(kg.d))和生理盐水,给药后24h,采血计数外周血淋巴细胞数目。结果给药前、第1次给药后2、4、8、24、48h外周血平均淋巴细胞数目分别为(9.575±1.1121)×109、(6.525±1.0175)×109、(3.512±1.0171)×109、(3.576±1.0046)×109、(3.513±1.0582)×109、(3.500±0.9606)×109/L;生理盐水和不同剂量的FTY720(0.3、1、3、10mg/kg)组的外周血平均淋巴细胞数目分别为(9.475±1.1121)×109、(7.825±1.3092)×109、(6.250±1.0821)×109、(5.100±0.9501)×109、(3.325±0.9977)×109/L。结论FTY720诱导外周血淋巴细胞数目减少有时间和剂量依赖性。给予FTY720(10mg/(kg.d))能够明显降低外周血淋巴细胞数目,并于用药后2h下降明显,用药后4h下降到谷值,4h以后维持于较低水平。外周血淋巴细胞数目和FTY720的剂量成直线关系,随FTY720剂量的增加而下降。
Objective To further investigate the effect of FTY720 on the reduction of peripheral blood lymphocytes in relation to dose and duration of administration. Methods FTY72010mg / (kg · d) was given to the mice. The number of peripheral blood lymphocytes was counted at 2,4,8,24,48h after the first administration and at different doses of FTY720 (0.3,1,3, 10mg / (kg.d)) and saline, 24h after administration, blood count the number of peripheral blood lymphocytes. Results Before treatment, the average numbers of peripheral blood lymphocytes in the 2nd, 4th, 8th, 24th and 48th hour after the first administration were (9.575 ± 1.1121) × 109, (6.525 ± 1.0175) × 109, (3.512 ± 1.0171) × (3.576 ± 1.0046) × 109, (3.513 ± 1.0582) × 109 and (3.500 ± 0.9606) × 109 / L, respectively. The peripheral blood of the rats in the normal saline and different doses of FTY720 (0.3,1,3,10 mg / kg) The average number of lymphocytes were (9.475 ± 1.1121) × 109, (7.825 ± 1.3092) × 109, (6.250 ± 1.0821) × 109, (5.100 ± 0.9501) × 109, (3.325 ± 0.9977) × 109 / L, respectively. Conclusion FTY720 induced a decrease in the number of peripheral blood lymphocytes in a time-and dose-dependent manner. Administration of FTY720 (10 mg / (kg · d)) significantly reduced the number of peripheral blood lymphocytes and decreased significantly at 2 h after administration. The level dropped to the trough at 4 h after administration and remained at a low level after 4 h. There was a linear relationship between the number of peripheral blood lymphocytes and the dose of FTY720, with the increase of FTY720 dose.
