目的:考察参蛭疏血滴丸对大鼠局灶性脑缺血模型的影响。方法:采用线栓法阻塞大脑中动脉制备大鼠局灶性脑缺血模型,将造模成功后48只大鼠随机分为假手术组、模型对照组、阳性对照组(血塞通片0.04 g.kg-1)和参蛭疏血滴丸高、中、低剂量组(0.936,0.468,0.234 g.kg-1),每组8只,各组均ig给药,1次/d,给药体积10 mL.kg-1,连续给药14 d。测定神经行为学评分、脑梗死面积比及血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。结果:与模型对照组比较,参蛭疏血滴丸高、中剂量组、阳性对照组可降低大鼠神经行为学评分,缩小脑梗死面积比、SOD活性明显升高(P<0.05,P<0.01),参蛭疏血滴丸高、中、低剂量组、阳性对照组MDA含量明显降低,(P<0.05,P<0.01)。结论:参蛭疏血滴丸对大鼠局灶性脑缺血有保护作用,其作用机制可能与抗自由基有关。
Objective: To investigate the effect of Shenzhi Shuxie Dropping Pills on focal cerebral ischemia in rats. Methods: The model of focal cerebral ischemia was established by occlusion of the middle cerebral artery by thread occlusion method. Forty rats were randomly divided into sham operation group, model control group and positive control group (Xuesaitong 0.04 g.kg-1) and Shenzhi Shuibixi dripping pills in high, middle and low dose groups (0.936, 0.468, 0.234 g.kg-1) Administration volume 10 mL.kg-1, continuous administration 14 d. Neurobehavioral score, infarct area ratio, serum SOD activity and malondialdehyde (MDA) content were determined. Results: Compared with the model control group, Shenzhi Shuxie Dropping Pill high, middle dose group and positive control group could reduce neurobehavioral score, reduce infarct area ratio and increase SOD activity (P <0.05, P < 0.01). The content of MDA in Shenjianshuxue Dropping Pills in high, middle and low dose group and positive control group was significantly lower than that in control group (P <0.05, P <0.01). Conclusion: Shenzhi Shuxie Dropping Pill has a protective effect on focal cerebral ischemia in rats, and its mechanism may be related to anti-free radical.
