目的探讨宫颈上皮内瘤变(CIN)及宫颈鳞状细胞癌中缺氧诱导因子-1α(HIF-1α)、葡萄糖转运蛋白-1(Glut-1)的表达及其与肿瘤生物学行为的关系和意义。方法收集2008年至2013年在如皋市人民医院手术的110例宫颈病变(48例CIN、62例宫颈鳞状细胞癌)组织及30例正常宫颈组织应用SP免疫组化法检测HIF-1α和Glut-1蛋白在不同宫颈组织中的表达并分析其相关性。结果 CIN、宫颈癌组HIF-1α和Glut-1阳性表达率均明显高于正常宫颈组(P均<0.01)。HIF-1α表达与临床分期及病理分级有关(P均<0.01)。Glut-1表达与肿瘤直径相关(P<0.01)。宫颈癌组织中HIF-1α、Glut-1的表达呈正相关(r=0.482,P<0.05)。结论宫颈鳞状细胞癌中HIF-1α和Glut-1蛋白的表达与肿瘤的恶性进展密切相关,联合检测HIF-1α和Glut-1可作为了解宫颈癌生物学行为的参考指标,并有可能为宫颈癌的基因治疗提供新的靶点和新的思路。
Objective To investigate the expression of HIF-1α and Glut-1 in cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma and its relationship with tumor biological behavior And meaning. Methods Totally 110 cases of cervical lesions (48 cases of CIN and 62 cases of cervical squamous cell carcinoma) and 30 cases of normal cervical tissues were collected from Rugao People’s Hospital from 2008 to 2013. SP immunohistochemistry was used to detect the expression of HIF-1α and Glut -1 protein in different cervical tissues and analyze their correlation. Results The positive rates of HIF-1α and Glut-1 in CIN and cervical cancer group were significantly higher than those in normal cervix group (all P <0.01). The expression of HIF-1α was correlated with clinical stage and pathological grade (all P <0.01). Glut-1 expression correlated with tumor diameter (P <0.01). The expression of HIF-1α and Glut-1 in cervical cancer tissues was positively correlated (r = 0.482, P <0.05). Conclusion The expression of HIF-1α and Glut-1 protein in cervical squamous cell carcinoma is closely related to the malignant progression of cervical squamous cell carcinoma. Combined detection of HIF-1α and Glut-1 may serve as a reference index for understanding the biological behavior of cervical cancer and may be Cervical cancer gene therapy to provide new targets and new ideas.
