目的探讨颈淋巴结结核患者外周血和组织中T-box基因家族转录因子(T-bet)的表达,并分析在结核病发病中的意义。方法采用实时荧光定量PCR(Realtime-PCR)检测了12例颈淋巴结结核患者和11例健康对照外周血T-bet的表达情况,使用GraphPad Prism 4.0统计软件进行分析,两组间比较采用配对t检验,以P<0.05为差异有统计学意义;同时采用免疫组织化学双染法检测颈淋巴结结核患者淋巴结组织中T-bet的表达情况,计数每个高倍视野中CD4+细胞和CD20+B细胞T-bet阳性细胞的百分率。结果在颈淋巴结结核患者外周血PBMC中T-bet的表达水平为6.07±3.35显著低于健康对照20.12±11.31(t=4.118,P=0.0005);淋巴结核结节中,CD4+细胞T-bet阳性率为65.7%,CD20+B细胞T-bet阳性率为5.0%。结论 T-bet在颈淋巴结结核外周血中低表达,而在淋巴组织中高表达,提示T-bet在颈淋巴结结核免疫发病中起重要的作用。
Objective To investigate the expression of T-box family transcription factors (T-bet) in peripheral blood and tissues of patients with cervical lymph node tuberculosis and to analyze its significance in the pathogenesis of tuberculosis. Methods Real-time quantitative PCR (Realtime-PCR) was used to detect the expression of T-bet in peripheral blood of 12 patients with cervical lymph node and 11 healthy controls. The data were analyzed by GraphPad Prism 4.0 software. The paired t- , P <0.05 for the difference was statistically significant; at the same time by immunohistochemical double staining of cervical lymph node tuberculosis tissue T-bet expression, count each high power field and CD20 + B cells T- bet positive cells. Results The expression level of T-bet in peripheral blood PBMC of patients with cervical lymph node tuberculosis was 6.07 ± 3.35, which was significantly lower than that of healthy controls (20.12 ± 11.31, t = 4.118, P = 0.0005) The rate of T-bet in CD20 + B cells was 65.7%. Conclusion T-bet is low expressed in the peripheral blood of cervical lymph node tuberculosis, but highly expressed in the lymphoid tissue, suggesting that T-bet plays an important role in the pathogenesis of tuberculosis immunodeficiency.
