目的:观察姜黄素(curcumine)对过氧化氢诱导的大鼠心肌细胞MCP-1表达的影响,探讨姜黄素在心肌细胞氧化应激中作用的可能机制。方法:以H2O2处理心肌细胞,构建心肌氧化损伤模型。并用姜黄素及H2O2共孵育心肌细胞,采用免疫印迹(Western blot)检测干预后不同时间MCP-1蛋白表达。结果:在干预后4、8、12、24、48h,H2O2组心肌细胞表达的MCP-1明显高于空白对照组(P<0.05),加入姜黄素共孵育后,各个时间点MCP-1的表达都明显下降(P<0.05)。结论:H2O2能诱导心肌细胞MCP-1蛋白表达,而姜黄素对H2O2诱导的心肌细胞MCP-1表达具有抑制作用,可能是其抗氧化应激的机制之一。
Objective: To observe the effect of curcumine on the expression of MCP-1 in cardiomyocytes induced by hydrogen peroxide and to explore the possible mechanism of curcumin in cardiomyocyte oxidative stress. Methods: Myocardial cells were treated with H2O2 to establish a model of myocardial oxidative damage. Cardiac myocytes were incubated with curcumin and H2O2. Western blot was used to detect the expression of MCP-1 at different time points after intervention. Results: MCP-1 expression in cardiomyocytes of H2O2 group was significantly higher than that of blank control group at 4, 8, 12, 24 and 48 h after intervention (P <0.05). After adding curcumin, MCP-1 Expression were significantly decreased (P <0.05). CONCLUSION: H2O2 can induce the expression of MCP-1 in cardiomyocytes, while curcumin can inhibit the expression of MCP-1 in H2O2-induced cardiomyocytes, which may be one of the mechanisms of its anti-oxidative stress.
