本文应用免疫金银染色(IGSS)法和图象定量分析法(TAS),对大鼠心肌细胞中雌激素受体(ER)作了定性、定位及相对含量的分析,并观察了心肌梗塞后大鼠心肌细胞ER的变化。结果表明:心肌细胞中存在有ER。ER在心脏中分布广泛,心室中ER密度大于心房中ER密度,两者分别为(0.2072±0.006;0.1687±0.005,P<0.01)。心肌梗塞(MI)后,坏死的心肌组织中无ER存在,非梗塞区ER密度增高,心房与心室分别为(0.2469±0.053;0.2719±0.006)与正常对照比较有显著差异。给予雌二醇处理后,心肌中ER密度降低,心房心室分别为(0.1276±0.003;0.1304±0.005),与前组比较均有显著差异。
In this study, the qualitative, localization and relative content of estrogen receptor (ER) in cardiomyocytes of rats were analyzed by immunogold-silver staining (IGSS) and image quantitative analysis (TAS) Changes of ER in rat cardiac. The results show that there is ER in cardiomyocytes. ER was widely distributed in the heart, and the density of ER in the ventricles was higher than that in the atrium (0.2072 ± 0.006; 0.1687 ± 0.005, P <0.01), respectively. Myocardial infarction (MI), no necrosis of myocardial tissue ER, non-infarction area ER density increased, atrial and ventricular were (0.2469 ± 0.053; 0.2719 ± 0.006) compared with the normal control were significantly different. After administration of estradiol, the density of myocardial ER decreased, atrial ventricle were (0.1276 ± 0.003; 0.1304 ± 0.005), respectively, which were significantly different from the former group.
